> Epitalon (AEDG) is a synthetic tetrapeptide modeled on the bovine pineal extract epithalamin, developed by Khavinson's St. Petersburg group. It activates telomerase and extends telomeres ~33% in human cell lines, with 2025 work adding a second route via ALT; animal lifespan data is positive. Human data is limited to two small, single-site Russian trials, retinitis pigmentosa (n=162) and circadian measures (n=75), with no Western replication.

> **Aliases:** Epithalon, Epithalone, Epithalamin (parent extract) **Sequence:** Ala-Glu-Asp-Gly (AEDG) — tetrapeptide **Molecular Weight:** ~390 Da (free base); salts commonly acetate or trifluoroacetate (TFA) **Origin:** Synthetic tetrapeptide based on amino acid composition of Epithalamin (bovine pineal gland extract). AEDG was confirmed in **authentic pineal polypeptide complex solution in 2017** (Khavinson et al., PMID [29124531](https://pubmed.ncbi.nlm.nih.gov/29124531/)) — it is endogenous, not purely a drug invention.

## Overview

Epitalon is a tetrapeptide (Ala-Glu-Asp-Gly) synthesized by **Vladimir Khavinson** and his team at the St. Petersburg Institute of Bioregulation and Gerontology based on the amino acid composition of **Epithalamin** , a polypeptide extract from the bovine pineal gland. It is one of the most studied anti-aging peptides, with research spanning over **25 years** since its 2001 Russian patent (referenced in [PMC11943447](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC11943447/)). It occupies the unique position of being a Russian Federation peptide bioregulator (short regulatory peptide class).

Epitalon's most celebrated property is its ability to **activate telomerase** and **elongate telomeres** — the protective caps on chromosomes that shorten with each cell division. This positions it as a potential **geroprotector** at the most fundamental cellular level.

**Evidence Level (as of Sept 2026):** Moderate-to-strong preclinical; modest human evidence. Key 2025 mechanistic advance: **dual mechanism** confirmed — telomerase upregulation AND Alternative Lengthening of Telomeres (ALT) activation (Igarashi et al., _Biogerontology_ 2025, **PMID[40908429](https://pubmed.ncbi.nlm.nih.gov/40908429/) / [PMC12411320](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12411320/)**). Telomerase activity alone is now known to be insufficient in many human cell lines; ALT adds a parallel telomere-maintenance route.

## Mechanism of Action

### Telomerase Activation & ALT (Updated 2025)

  * **Reactivates hTERT expression** in telomerase-negative human fetal fibroblast cultures (Khavinson, Bondarev, Butyugov, 2003, PMID [12937682](https://pubmed.ncbi.nlm.nih.gov/12937682/))
  * **Elongates telomeres** by an average of **33.3%** in human somatic cells
  * **2025 update — ALT pathway also engaged:** Igarashi S et al., _Biogerontology_ 2025 Sep 4;26(5):178 (**[PMC12411320](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12411320/)**). Demonstrated Epitalon increases telomere length in human cell lines through **either** telomerase upregulation **or** Alternative Lengthening of Telomeres (ALT) activity, depending on baseline hTERT status. This is a mechanism-resolving finding: it explains how Epitalon can act on ALT-positive tumors as well as telomerase-reactivated somatic cells (and supports its reported anti-tumor profile).
  * Control fibroblasts lost mitotic capacity after passage 34; Epitalon-treated cells continued past passage 44 (Khavinson, May 2004 _Bull Exp Biol Med_).
  * Telomere repeat amplification protocol (TRAP) confirms telomerase induction in HeLa and 602/17 fetal fibroblasts.

### Pineal Gland & Melatonin Axis

  * **Restores melatonin synthesis** in aged pinealocytes by elevating AANAT and pCREB expression (more potent at 3h than Vilon; PMID [22816096](https://pubmed.ncbi.nlm.nih.gov/22816096/), Khavinson 2012)
  * **In vivo in old rhesus monkeys (22.8 ± 1 yr):** IM Epitalon normalized cortisol and stimulated melatonin synthesis in age-dependent manner (Goncharova et al.)
  * **In vitro conflict:** Djeridane et al. 2003 (PMID [12809170](https://pubmed.ncbi.nlm.nih.gov/12809170/)) found **no** direct melatonin secretion effect in isolated perifused pineal glands from young/old rats — suggesting Epitalon acts on synthesis machinery rather than secretion per se
  * **Human circadian RCT signal:** 75 women, 0.5 mg/day sublingual × 20 days → 1.6× urinary 6-sulfatoxymelatonin (aMT6s) vs placebo; Clock gene expression −1.8×, Cry2 +2×, Csnk1e −2.1× (p < 0.05). Mechanistic support for geroprotective activity via clock gene modulation.

### Gene Regulation & DNA Binding

  * **Direct DNA binding** confirmed (FITC-labeled Epitalon penetrates HeLa cells and reaches the nucleus; binds CAG-repeat sequences susceptible to cytosine methylation)
  * In silico: binds ATTTG and ATTTC promoter sequences — the **ATTTC motif is repeated multiple times in the telomerase gene promoter** (Khavinson 2004)
  * **Epigenetic regulation:** interacts with histone H1/3 and H1/6 (binding energies −56.49 and −64.51 kcal/mol; molecular dynamics AMBER12EHT) — first non-DNA epigenetic route
  * **Plant system validation:** _Nicotiana tabacum_ callus assays confirm Epitalon binds CAG regions of plant DNA
  * Solovyev et al.: Epitalon reduces dsDNA melting temperature by up to **41 °C** at 0.1 M NaCl ionic strength

### Transporter Binding (New 2020 finding)

  * **In silico docking:** strong binding to LAT1, LAT2, PEPT1, PEPT2 amino acid transporters (comparable to known PEPT1 inhibitors). Suggests **anti-cancer mechanism via amino acid transport blockade** — explains observed tumor-suppression without direct cytotoxicity.

### Antioxidant & Nrf2

  * **Direct Keap1/Nrf2 promoter binding** demonstrated (Gutop et al.) — upregulates SOD-1, NQO1, catalase expression
  * In _D. melanogaster_ , 16,000-fold lower concentration than melatonin achieves comparable lifespan extension — "signaling role" rather than stoichiometric antioxidant

### Other Mechanisms

  * **STAT1 phosphorylation** in THP-1 monocytic leukemia cells (receptor-independent; additive with LPS → ERK1/2 phosphorylation; **suppresses STAT3**)
  * **Cholinergic:** +10–25% AChE/BuChE activity in SH-SY5Y (Nalieva et al.); −20% soluble APP secretion → potential AD-relevant modulation
  * **Anti-mutagenic** in albino & grey mice (sperm head abnormalities, micronuclei in peripheral blood erythrocytes)
  * **Anti-apoptotic:** 2.12-fold reduction in splenic lymphocyte apoptosis post-γ-irradiation (Kozina)
  * **Cytokine modulation:** IL-2 mRNA elevation in splenocytes (5h vs 20h for Vilon); regioselective hypothalamic IL-2 mRNA changes after IM/intranasal

### Anti-Tumor Effects

  * **HER-2/neu transgenic FVB/N mice:** 3.7-fold lower HER-2/neu mRNA expression; reduced mammary adenocarcinoma count (p < 0.05)
  * **C3H/He mice:** 6.5-month study — reduced tumor incidence, multiplicity, localization vs saline
  * **SAMP-1 mice (accelerated aging model):** Epitalon reduced chromosomal abnormalities more than melatonin in SHR controls
  * **1,2-dimethylhydrazine colon carcinogenesis model:** significant decrease in cancer incidence
  * **Frailty-model dependent:** Semenchenko et al. showed Epitalon **either** reduced max lifespan (debilitation/frailty accumulation) **or** enhanced survival (robustness/heterogeneity) depending on dose schedule — non-monotonic dose-response

## 2025 Mechanism Update: Dual Telomere Maintenance

The single most significant mechanistic advance for Epitalon in 2025 was the demonstration that **Epitalon engages both the telomerase AND ALT pathways** depending on the baseline hTERT status of the cell (Igarashi S et al., _Biogerontology_ 2025 Sep 4;26(5):178, **PMID[40908429](https://pubmed.ncbi.nlm.nih.gov/40908429/) / [PMC12411320](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC12411320/)**). This:

  1. Resolves the long-standing puzzle of how a single tetrapeptide produces effects in ALT-positive (telomerase-negative) tumor cells as well as in telomerase-reactivated somatic cells.
  2. Confirms Epitalon has activity independent of canonical hTERT induction — broader applicability than previously assumed.
  3. Aligns with the documented anti-tumor profile (anti-cancer effect via amino acid transporter blockade is mechanistically independent of telomere status).

Companion 2025 finding (Ullah S et al., _Life Sci_ 2025;362:123381, cited in Araj review): Epitalon **enhanced bovine oocyte maturation and post-thawed embryo development** via telomerase + mitochondrial pathways (PGC-1α, Sirt-1, tFAM, BCL2 ↑; JC-1 mito health ↑; intracellular ROS ↓). First evidence that Epitalon accelerates blastocyst expansion/hatching — relevant to reproductive longevity research.

## Doses in studies

> ⚠️ **Disclaimer:** Research protocols. Epitalon is not FDA-approved. Quality concerns: **Vanhee et al.** confirmed LC-MS/MS is the only reliable way to distinguish genuine AEDG from EADG (Glu-Ala-Asp-Gly) counterfeit, which arises via N-terminal glutamic acid pyroglutamate formation during heat treatment. Counterfeit risk is non-trivial in unregulated gray market.

### Approaches Reported in the Literature

  * **Often combined with** Thymogen (Glu-Trut dipeptide) for immune support or [other pineal peptides](peptide-bioregulation-khavinson.html)
  * **Retinal protocol** (per Russian clinical trial, St. Petersburg IBG): 5 µg/eye parabulbar × 10 days

## Safety Profile

Aspect | Assessment
---|---
**Overall safety** | Excellent — no reported toxic, allergic, or adverse effects across 40+ years of Russian clinical use
**Acute toxicity** | None observed even at high doses in animal studies
**Long-term safety** | Used clinically in Russia since the 1990s
**Cancer risk** | **Anti-tumor** rather than pro-tumor — reduces tumor incidence in animal models across multiple strains
**FDA status** | Not approved in the United States; **no IND/CTA in US** ; subject of 2026 PCAC compounding panel review (see [FDA Peptide Reclassification 2026 — Regulatory Intelligence](fda-peptide-reclassification-2026.html))
**Russian clinical experience** | n=162 retinitis pigmentosa patients; no side effects reported
**Stability concern** | Short peptide; rapid in vivo degradation. Authors of [PMC11943447](https://www.ncbi.nlm.nih.gov/pmc/articles/PMC11943447/) recommend acetylation/amidation modification as future direction — **not yet evaluated**
**Stereoisomer question** | 8 possible stereoisomers; only L-amino acid form characterized. Other 7 unstudied
**Counterfeit risk** | Significant in gray market (EADG vs AEDG confusion, pyroglutamate formation); Vanhee LC-MS/MS is the gold standard

## Clinical Reality Check (Sept 2026)

  * **No FDA-approved indication anywhere outside Russia**
  * **Clinical trial landscape:** All published human data is Russian (St. Petersburg IBG, Goncharova lab). Two human signals: retinitis pigmentosa (n=162, parabulbar) and circadian (n=75 women, sublingual). Both small, single-site, no Western replication.
  * **Mechanism is now better understood** thanks to 2025 Igarashi ALT paper + transporter docking
  * **Quality control is the active risk:** LC-MS/MS confirmation per Vanhee et al.; the EADG counterfeit is indistinguishable by mass alone
  * **Stability is an unresolved pharmacokinetic challenge** — no published acetylated/amidated analog studies
